ABSTRACT

Human alveolar echinococcosis is increasingly documented in Alberta, Canada. Its causative agent, Echinococcus multilocularis (Em), can be transmitted to humans by infected dogs. We assessed the prevalence and associated risk factors for Em infections in domestic dogs in Calgary, Alberta, Canada. In this cross-sectional study that coupled collection and assessment of dog feces with a survey on potential risk factors, 13 of 696 (Bayesian true prevalence, 2.4%; 95% CrI: 1.3-4.0%) individual dogs' feces collected during August and September 2012 were qPCR positive for Em. Sequencing two of these cases indicated that both were from the same Em European strain responsible for human infections in Alberta. Likelihood of intestinal Em was 5.6-times higher in hounds than other breeds, 4.6-times higher in dogs leashed at dog parks than those allowed off-leash, 3.1-times higher in dogs often kept in the backyard during spring and summer months than those rarely in the yard, and 3.3-times higher in dogs living in neighbourhoods bordering Bowmont park than those in other areas of Calgary. This situation warrants surveillance of dog infections as a preventative measure to reduce infections in North America.

ABSTRACT

Giardia spp. and Cryptosporidium spp. are common gastrointestinal parasites with the potential for zoonotic transmission. This study aimed to (1) determine the genotypes occurring in dogs and coyotes occupying a similar urban area; (2) determine if these hosts were infected with potentially zoonotic genotypes; (3) provide baseline molecular data. In August and September 2012, 860 dog owners living in neighborhoods bordering six urban parks in Calgary, Alberta, Canada, provided faecal samples from their dogs. From March 2012 through July 2013, 193 coyote faeces were also collected from five of six of the same parks. Direct immunofluorescence microscopy (DFA) indicated that Giardia spp. and Cryptosporidium spp. infected a total of 64 (7.4%) and 21 (2.4%) dogs, as well as 15 (7.8%) and three (1.6%) coyotes, respectively. Semi-nested, polymerase chain reactions targeting the 16S small-subunit ribosomal ribonucleic acid (SSU rRNA) and 18S SSU rRNA genes of Giardia spp. and Cryptosporidium spp., respectively, were conducted on samples that screened positive by DFA, and products were sequenced and genotyped. Dogs were infected with Giardia intestinalis canid-associated assemblages C (n = 14), D (n = 13), and Cryptosporidium canis (n = 3). Similarly, G. intestinalis assemblages C (n = 1), D (n = 1) and C. canis (n = 1), were detected in coyotes, as well as G. intestinalis assemblage A (n = 1) and Cryptosporidium vole genotype (n = 1). Dogs and coyotes were predominantly infected with host-specific genotypes and few potentially zoonotic genotypes, suggesting that they may not represent a significant risk for zoonotic transmission of these parasites in urban areas where these hosts are sympatric.

ABSTRACT

BACKGROUND: A community-wide outbreak of Legionnaires' disease (LD) occurred in Genesee County, Michigan, in 2014 and 2015. Previous reports about the outbreak are conflicting and have associated the outbreak with a change of water source in the city of Flint and, alternatively, to a Flint hospital. OBJECTIVE: The objective of this investigation was to independently identify relevant sources of Legionella pneumophila that likely resulted in the outbreak. METHODS: An independent, retrospective investigation of the outbreak was conducted, making use of public health, health care, and environmental data and whole-genome multilocus sequence typing (wgMLST) of clinical and environmental isolates. RESULTS: Strong evidence was found for a hospital-associated outbreak in both 2014 and 2015: a) 49% of cases had prior exposure to Flint hospital A, significantly higher than expected from Medicare admissions; b) hospital plumbing contained high levels of L. pneumophila; c) Legionella control measures in hospital plumbing aligned with subsidence of hospital A-associated cases; and d) wgMLST showed Legionella isolates from cases exposed to hospital A and from hospital plumbing to be highly similar. Multivariate analysis showed an increased risk of LD in 2014 for people residing in a home that received Flint water or was located in proximity to several Flint cooling towers. DISCUSSION: This is the first LD outbreak in the United States with evidence for three sources (in 2014): a) exposure to hospital A, b) receiving Flint water at home, and c) residential proximity to cooling towers; however, for 2015, evidence points to hospital A only. Each source could be associated with only a proportion of cases. A focus on a single source may have delayed recognition and remediation of other significant sources of L. pneumophila. https://doi.org/10.1289/EHP5663.

ABSTRACT

Associations between park use and infections with gastrointestinal (GI) parasites in dogs (Canis familiaris) have been previously observed, suggesting park use may pose risks for infection in dogs, and potentially, in humans. This study was conducted to establish the overall level of perceived risk of parasitism in dogs, the frequency of unleashing dogs in parks, and to determine if dog owners' risk perceptions of parasite transmission among humans and dogs are associated with the reported frequency of unleashing dogs. From June to September 2010, 635 surveys were administered to dog owners in nine city parks in Calgary, Alberta, by the lead author to explore dog-walking behaviors in parks under differing leashing regulations. From these, a subset of 316 questionnaires were analyzed to examine associations between behavioral and dog demographic factors, risk perception and acceptability of perceived risks of dog and human parasitism, and education regarding parasitism in dogs and humans. Multivariate statistics were conducted using three separate Chi-Square Automatic Interaction Detection (CHAID) decision trees to model risk perception of dogs becoming parasitized while in the park, risk perception of zoonotic transmission, and off-leash frequency. Predictors included recreational behaviors, dog demographics, risk perception of park-based and zoonotic transmission, education regarding parasites, and leashing regulations (e.g. on-leash, off-leash, or mixed management parks). The perceived risk of park-based transmission was relatively higher than perception of zoonotic transmission and the majority of people unleashed their dogs at least some of the time. Risk perception was not associated with off-leash frequency in dogs and risk perception and off-leash frequency were associated with factors other than each other. The results suggest owners may underestimate the potential risks for parasitism related to some dog-walking behaviours, and are relevant for public and animal health.

BACKGROUND: In urban parks, dogs, wildlife and humans can be sympatric, introducing the potential for inter- and intra-specific transmission of pathogens among hosts. This study was conducted to determine the prevalence of zoonotic and non-zoonotic gastrointestinal parasites in dogs in Calgary city parks, and assess if dog-walking behaviour, park management, history of veterinary care, and dog demographics were associated with parasitism in dogs METHODS: From June to September 2010, 645 questionnaires were administered to dog owners in nine city parks to determine behavioural and demographic factors, and corresponding feces from 355 dogs were collected. Dog feces were analyzed for helminth and some protozoan species using a modified sugar flotation technique and microscopic examination, a subsample was analyzed for Giardia spp. and Cryptosporidium spp. using a direct immunofluorescence assay. Descriptive and multivariate statistics were conducted to determine associations among behaviours, demographics, and parasite prevalence and infection intensities RESULTS: Parasite prevalence was 50.2%. Giardia spp. (24.7%), Cryptosporidium spp. (14.7%), and Cystoisospora spp. (16.8%) were the most prevalent parasites. Helminth prevalence was low (4.1%). Presence of Giardia spp. was more likely in intact and young dogs; and infection with any parasite and Giardia spp. intensity were both positively associated with dogs visiting multiple parks coupled with a high frequency of park use and off-leash activity, and with being intact and young. Cryptosporidium spp. intensity was associated with being intact and young, and having visited the veterinarian within the previous year CONCLUSIONS: Our results indicate a higher overall prevalence of protozoa in dogs than previously found in Calgary. The zoonotic potential of some parasites found in park-attending dogs may be of interest for public health. These results are relevant for informing park managers, the public health sector, and veterinarians.
